A Comparison of Estimates of Michaelis-Menten Kinetic Constants from Various Linear Transformations

چکیده
The parameters which characterize this equation, and which must ordinarily be estimated from the observed data, are ‘v,,,, the maximum initial velocity which is theoretically attained when the enzyme has been “saturated” by an infinite concentration of substrate, and K,, the Michaelis constant which is numerically equal to the concentration of substrate for half-maximal initial velocity. Since the relationship between the independent variable, Cs, and the dependent variable, v, is curvilinear, it has long been customary to facilitate estimation of the two parameters by plotting the experimental data according to one of the following three linear transformations of Equation 1.

متن کاملDetermination of Kinetic Parameters for Catalytic Isomerization of Glucose to Fructose by immobilized Glucose Isomerase in an Aqueous-Ethanol Medium
Catalytic isomerization of glucose to fructose by immobilized glucose isomerase in an aqueous-ethanol medium was studied. Using Michaelis-Menten equation and Haldane relationship, the main kinetic parameters were determined: Kmf, Vmf, Kmr, Vmr. By comparing the aqueous-ethanol medium with an aqueous solution, the measured values of Kmr and Kmf were increased in the aqueous-ethanol solution, whi...

متن کاملThe Kinetics of Enzyme Mixtures
Even purified enzyme preparations are often heterogeneous. For example, preparations of aspartate aminotransferase or cytochrome oxidase can consist of several different forms of the enzyme. For this reason we consider how different the kinetics of the reactions catalysed by a mixture of forms of an enzyme must be to provide some indication of the characteristics of the species present. Based...
متن کاملThe direct linear plot. A new graphical procedure for estimating enzyme kinetic parameters.
A new plot is described for analysing the results of kinetic experiments in which the Michaelis-Menten equation is obeyed. Observations are plotted as lines in parameter space, instead of points in observation space. With appropriate modifications the plot is applicable to most problems of interest to the enzyme kineticist. It has the following advantages over traditional methods of plotting ki...
